Multiple copies of the gene of interest in synthesized in vitro using DNA polymerase enzyme.
1. Which are the three steps of the above process?
2. Find out the DNA polymerase enzyme used here.
Answer:
1. Three steps of the above process are:
- Denaturation – The target DNA is heated to a high temperature (about 94°C). It results in separation of the two strands. Each single strand of the target DNA acts as a template for DNA synthesis.
- Annealing – The two oligonucleotide primers anneal to each of the single-stranded template DNA at it’s 3’ end. Annealing occurs at low temperature (40°C – 60°C)
- Extension – DNA polymerase enzyme extends the primers by adding nucleotides complementary to ) the template provided in the reaction.
2. Thermostable DNA polymerase/Taq polymerase.
